Role of Fatty Aldehyde Dehydrogenase in Ether Lipid Degradation Pathways

چکیده
Ether Lipid Metabolism Ether-linked glycerolipids are present in most mammalian tissues including brain, kidney, heart and skin. They were once strictly referred to as structural components of membranes, but by now it is known that some are biologically active (1). Ether lipids have as important roles as maintaining proper brain function, spermatogenesis, mediation of inflammation and lens organisation (2-4). The ether bond of ether lipids is synthesised by the action of two peroxisomal enzymes, dihydroxyacetonephosphate acyltransferase (DHAPAT) and alkyl-dihydroxyacetonephosphate synthase (ADAPS) (5, 6). Lack of ether lipids in DHAPAT knockout mice causes early arrest of spermatogenesis, formation of a lenticonus anterior accompanied with the generation of cataract and hypomyelination in distinct regions of the central nerve system (reviewed by 2).

The role of fatty aldehyde dehydrogenase in epidermal structure and function
The epidermal water barrier resides in the stratum corneum (SC) and is dependent on a highly organized network of multi-lamellar membranes comprised of a critical lipid composition. The SC membranes are formed from precursor membranes packaged in cytoplasmic lamellar bodies in the stratum granulosum and delivered to the SC by exocytosis. An abnormal lipid composition of the SC membranes often r...

The mechanism of synthesis of plasmalogens is not currently known, but the incorporation of labeled phosphate, acetate, fatty acids, and fatty aldehydes into plasmalogens has been described for intact systemsl-B. Such isotopic studies have suggested a variety of pathways leading to the synthesis of plasmalogens. Among these proposed pathways are the dehydrogenation of the alkyl ether bond, the ...
متن کاملChlorinated lipid species in activated human neutrophils: lipid metabolites of 2-chlorohexadecanal.
Neutrophils are important in the host response against invading pathogens. One chemical defense mechanism employed by neutrophils involves the production of myeloperoxidase (MPO)-derived HOCl. 2-Chlorohexadecanal (2-ClHDA) is a naturally occurring lipid product of HOCl targeting the vinyl ether bond of plasmalogens. Previous studies have shown that exogenously-added 2-ClHDA is oxidized to 2-chl...
متن کاملStudy on the Regulatory Mechanism of the Lipid Metabolism Pathways during Chicken Male Germ Cell Differentiation Based on RNA-Seq
Here, we explore the regulatory mechanism of lipid metabolic signaling pathways and related genes during differentiation of male germ cells in chickens, with the hope that better understanding of these pathways may improve in vitro induction. Fluorescence-activated cell sorting was used to obtain highly purified cultures of embryonic stem cells (ESCs), primitive germ cells (PGCs), and spermatog...
متن کاملFatty Aldehydes in Cyanobacteria Are a Metabolically Flexible Precursor for a Diversity of Biofuel Products
We describe how pathway engineering can be used to convert a single intermediate derived from lipid biosynthesis, fatty aldehydes, into a variety of biofuel precursors including alkanes, free fatty acids and wax esters. In cyanobacteria, long-chain acyl-ACPs can be reduced to fatty aldehydes, and then decarbonylated to alkanes. We discovered a cyanobacteria class-3 aldehyde-dehydrogenase, AldE,...
